Oak Ridge National Laboratory researchers reported that more than 93% of possible E. coli CRISPR-Cas9 guide RNAs worked as intended after testing nearly 500,000 guides. The team also identified why a small fraction failed and developed a method to repair them. ORNL made the resulting gRNA Atlas available without charge as an interactive application for exploring potential Cas9 target sites across the bacterium's genome. The work was published in Nucleic Acids Research.
