IIT Gandhinagar team develops pyridinium fluorescent probes for plant xylem

The work, published in Plant and Cell Physiology (DOI: 10.1093/pcp/pcag084), introduces a faster, more specific and sensitive staining method than conventional dyes used on thin plant tissue slices. Standard stains such as propidium iodide, berberine, basic fuchsin, or rhodamine bind broadly to charged or aromatic compounds in cell walls.
The new class centers on pyridinium derivatives. A C1 pyridinium probe selectively highlights xylem in Arabidopsis thaliana tissue in images released with the study, contrasting with broader basic fuchsin staining.
The authors say higher sensitivity and specificity should make it easier to investigate plant development, vascular defects, and responses to environmental conditions. They note further work is needed to determine how the dyes bind the cell wall and to adapt them for living tissue so researchers can watch water-conducting vessels form in real time.
